Target Information
Perforin is a major cytolytic protein found in cytolytic granules. It functions as a pore-forming mediator involved in immune defense against tumors and viral infections, primarily through cytotoxic lymphocytes.
Perforin consists of 555 amino acids, including a 21 amino acid signal peptide, and has a molecular weight of 70–75 kDa.
It shares structural homology with complement component C9 and operates via transmembrane channel formation.
Expression is limited to killer cell lines, not helper T cells or other tumor cells.
Defects in the perforin gene are associated with familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder of early childhood.
Alternative splicing yields multiple transcript variants.
