Target Information
Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates specific proline residues in the oxygen-dependent degradation (ODD) domains of HIF1A and HIF2A, primarily at the CODD site. Hydroxylated HIFs are targeted for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. Under hypoxic conditions, hydroxylation is reduced, allowing HIFs to translocate to the nucleus and activate hypoxia-inducible genes.
EGLN3 (PHD3) also hydroxylates PKM, ADRB2, and TELO2, influencing glycolysis, apoptosis, and DNA damage response pathways. Plays a key role in regulating cardiomyocyte and neuronal apoptosis, and hypoxic inflammation.
