Target Information

LARP1 is an RNA-binding protein that regulates translation of specific mRNA species downstream of the mTORC1 complex, in response to growth signals and nutrient availability.
It interacts with both the 3′ poly-A tails of mRNAs and the 7-methylguanosine cap structure of mRNAs containing a 5′ terminal oligopyrimidine (5′TOP) motif, which includes mRNAs encoding ribosomal proteins and translation machinery components.
This interaction represses translation by preventing EIF4G1 binding. Upon mTORC1 activation, LARP1 is phosphorylated and dissociates from the 5′ UTR of mRNAs.
It associates with ribosomes and enhances translation of 5′TOP motif-containing mRNAs, promoting protein synthesis, cell growth, and proliferation.
LARP1 also stabilizes these mRNAs, preventing apoptosis.
