Target Information
SHP-1 (PTPN6)은 주로 조혈세포에서 발현되는 비수용체 단백질 티로신 인산화효소입니다. 이 효소는 두 개의 SH2 도메인, 티로신 인산화효소 촉매 도메인 및 카복시말단 조절 도메인으로 구성되어 있습니다. SHP-1은 타깃 단백질의 인산기를 제거하여 여러 티로신 키나아제 조절 경로를 하향 조절합니다. 조혈세포에서 SHP-1의 아미노 말단 SH2 도메인은 인산화된 에리트로포이에틴 수용체(EpoR)에 결합하여 조혈 성장의 음성 조절을 수행합니다. 상피세포에서 SHP-1의 과발현은 Ros 수용체 티로신 키나아제의 탈인산화를 유도하여 Ros 의존적 세포 증식 및 변형을 억제합니다. 골수세포에서 리간드 결합 후 SHP-1은 IL-3R 베타 사슬과 결합하여 IL-3 유도 티로신 인산화 및 세포 증식을 하향 조절합니다.
