Target Information
The Ragulator complex is composed of three proteins: C11orf59, ROBLD3, and MAPKSP1, which target mTORC1 to lysosomal surfaces. This endosomal trafficking process can be exploited by certain viruses, including coronaviruses, which show increased replication when C11orf59 is disrupted.
C11orf59 is regulated by cellular cholesterol levels and is involved in LDL uptake and cholesterol homeostasis. It interacts with PPAR-alpha to control lipid and carbohydrate metabolism in diabetes.
Nearly all effects of C11orf59 are associated with mTORC1 kinase growth promotion and dysregulation of the normal mTOR pathway.
