Target Information

Sclerostin is a glycoprotein predominantly secreted by osteocytes and a member of the Cerberus/DAN family of BMP antagonists. It functions as an endogenous regulator of the canonical Wnt signaling pathway and bone homeostasis. Although mainly expressed by osteocytes, sclerostin is also present in bone, bone marrow, cartilage, kidney, and liver, and is produced by hypertrophic chondrocytes and cementocytes.
Sclerostin interacts with LRP5, LRP6, and LRP4 to inhibit Wnt signaling and can promote osteoclast activity by increasing RANKL expression in osteocytes. Its physiological importance is highlighted by bone dysplasia disorders such as sclerosteosis and van Buchem disease, caused by mutations leading to reduced sclerostin levels and excessive bone formation.
