Product Specific Information

Description:
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 plays a key role in peripheral tolerance and autoimmune disease, predominantly expressed on activated T and B lymphocytes. PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC), which show overlapping functions.

Costaining experiments suggest eBioJ105 recognizes a different epitope than MIH4 (Product #11-9969).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on stimulated human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333). Clone-specific performance should be determined empirically.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

PD-1 (PDCD-1) regulates T-cell activation by delivering inhibitory signals upon binding to its ligands PD-L1 and PD-L2 on antigen-presenting cells. This mechanism prevents excessive immune activation. PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune diseases. PD-1 often migrates at higher molecular weight in SDS-PAGE despite its predicted size.
