Target Information

Ubiquitin-like protein ISG15 plays a key role in the innate immune response to viral infection, either via conjugation to target proteins (ISGylation) or as a free protein. It is secreted from monocytes in response to type I interferons and promotes NK-cell proliferation and non-MHC-restricted cytotoxicity.
ISG15 synthesis is stimulated by IFN-α, IFN-β, or IFN-ω, but not IFN-γ. Expression is also induced by overexpression of interferon regulatory factors and by influenza B virus.
ISG15 is secreted by monocytes, T-lymphocytes, B-lymphocytes, fibroblasts, and epithelial cells. Its activity in enhancing NK cell proliferation and inducing IFN-γ from T cells suggests a role in amplifying the immunomodulatory effects of interferons.
Mutations in the ISG15 gene can result in Immunodeficiency 38, with basal ganglia calcification (IMD38).
