Rabphilin-3A의 카복시 말단은 두 개의 C2 도메인(A와 B)을 포함하며, 아미노 말단(45–170번 잔기)은 두 개의 zinc finger 모티프를 가진 cysteine-rich 영역을 포함합니다. Rabphilin-3A는 synaptotagmins 및 Doc2를 포함하는 C-type tandem C2 단백질 계열에 속합니다.
Rabphilin은 신경내분비 세포에 발현되며 Rab3A와 함께 시냅스 소포 및 chromaffin 과립에서 공위치를 이룹니다. Rabphilin-3A는 Rab3a/GTP/Mg+와 결합하며, C2 도메인 내에서 칼슘 이온과 phosphatidylinositol 4,5-bisphosphate 함유 지질 소포와 결합합니다.
Rabphilin-3A는 칼슘 의존성 분비의 양성 조절자이며, Rab3a는 음성 조절자입니다. 두 단백질은 서로 독립적으로 분비를 조절하며, Rabphilin-3A의 작용은 Rab3 결합 도메인을 인식하는 미지의 인자에 의해 매개될 가능성이 있습니다.
