This antibody recognizes an extracellular/luminal epitope of CD107b (LAMP-2), an extensively glycosylated 100–120 kDa lysosome-associated protein widely expressed in human tissues.
Target Information
LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of the lysosomal membrane glycoprotein family. It provides carbohydrate ligands for selectins and may play roles in tumor cell metastasis, lysosomal maintenance, and adhesion.
Prior to posttranslational modification, LAMP2 is a ~46 kDa polypeptide. The mature form is glycosylated to a molecular weight of ~100–130 kDa. Structurally, LAMP2 consists of:
- A large amino-terminal intra-lysosomal domain
- A hydrophobic transmembrane domain
- A short carboxyl-terminal cytoplasmic tail
LAMP2 is used to monitor chaperone-mediated autophagy and exists in three variant forms: LAMP-2A, LAMP-2B, and LAMP-2C. Together with CD107a/LAMP-1, it forms a major component of lysosomal membranes, essential for lysosome-phagosome fusion and phagosomal maturation. LAMP2 deficiency results in autophagosome accumulation, leading to cardiomyopathy and myopathy (Danon’s disease).
