Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It serves as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and facilitating iron recycling through endocytosis and heme breakdown.

CD163 is expressed on human monocytes and macrophages, especially during the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, excluding certain lymphoid follicle cells and Langerhans cells. Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 into plasma or supernatant.

CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, with higher affinity for multimeric haptoglobin (HP1F phenotype). It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
