Background

The protein encoded by the GPX4 gene belongs to the glutathione peroxidase family, which catalyzes the reduction of hydrogen peroxide, organic hydroperoxides, and lipid hydroperoxides, thereby protecting cells against oxidative damage. This isozyme shows high preference for lipid hydroperoxides and plays a key role in preventing membrane lipid peroxidation and cell death. GPX4 is also essential for normal sperm development and functions as both a peroxidase and a structural protein in mature spermatozoa. Mutations in GPX4 are associated with Sedaghatian type of spondylometaphyseal dysplasia (SMDS). The protein is a selenoprotein containing the rare amino acid selenocysteine (Sec) at its active site, encoded by the UGA codon. The SECIS element in the 3' UTR of the mRNA enables recognition of UGA as a Sec codon. Alternatively spliced transcript variants have been identified for this gene. (RefSeq, Oct 2016)
