Recommended positive controls: rat brain, MDCK.

This antibody stains clathrin-coated membranes at the plasma membrane, and in the Golgi region of methanol-fixed MDBK cells. By Western bloy on bovine and rat brain preparations, this antibody detects a major band of AP-1 at ~115 kDa and a minor component at ~110 kDa, and bands at ~104 and ~112 kDa representing AP2. The antibody reacts with polypeptides at ~100 kDa in bovine liver, human heart fibroblasts and MDBK cells, but not with any components in the 110-115 kDa range from these sources, or from rat PC12 cells, neuroblastoma or astrocytes.

Target Information

Adaptor protein complex 1 is found at the cytoplasmic face of coated vesicles located at the Golgi complex, where it mediates both the recruitment of clathrin to the membrane and the recognition of sorting signals within the cytosolic tails of transmembrane receptors. This complex is a heterotetramer composed of two large, one medium, and one small adaptin subunit. The protein encoded by this gene serves as one of the large subunits of this complex and is a member of the adaptin protein family. This gene is a candidate meningioma gene. Alternative splicing results in multiple transcript variants.
