Target Information
ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration. The Rho/ROCK pathway is involved in the progression of various human cancers. ROCK1 has been identified as a Caspase-3 substrate.
ROCK1 contributes to myosin light chain (MLC) activation through two mechanisms: direct phosphorylation of MLC and phosphorylation-mediated inhibition of MLC phosphatase. This leads to increased cellular contractility, necessary for apoptotic membrane blebbing.
ROCK1 consists of an N-terminal kinase domain and a C-terminal inhibitory cysteine/histidine-rich domain within a pleckstrin-homology region. These domains are linked by a variable region containing the Rho-binding domain. During apoptosis, ROCK1 is cleaved by caspase-3 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity. This cleavage induces MLC phosphorylation and membrane blebbing, key events in apoptosis execution.
