Product Specific Information

The MIH5 monoclonal antibody reacts with mouse B7-H1 (PD-L1), a B7 family member with a predicted molecular weight of ~40 kDa. B7-H1 is expressed on T, B, NK, and dendritic cells and serves as a ligand for PD-1. The interaction of PD-1 with PD-L1 or PD-L2 inhibits T and B cell responses. MIH5 is reported as a blocking antibody.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Optimal concentration should be titrated empirically.

Super Bright 600 is a tandem dye excited by the 405 nm violet laser and emits at 600 nm (recommended 610/20 bandpass filter). Ensure instrument compatibility.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C with minimal signal loss.

Target Information

Programmed death receptor ligand 1 (PD-L1, also known as B7-H1) is a B7 family member that binds to PD-1, negatively regulating T-cell receptor (TCR) signaling. PD-L1 engagement reduces TCR-mediated proliferation and cytokine production. It is abundantly expressed on many murine and human cancers and upregulated by IFN-gamma, suggesting a role in tumor immune evasion.
