Product Specific Information

The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins. D7 recognizes both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells.
Sca-1 is expressed highly upon activation regardless of Ly-6 haplotype and serves as a major phenotypic marker for mouse hematopoietic progenitor/stem cell subsets.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number should be optimized empirically (10⁵–10⁸ cells/test).

Fluorophore Details:

• Super Bright 600 is a tandem dye excited by 405 nm violet laser, emitting at 600 nm.
• Recommended filter: 610/20 bandpass.
• Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright conjugates to minimize non-specific polymer interactions.
• Light-sensitive; protect from light.
• Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

Stem Cell Antigen-1 (Sca-1) is a member of the Ly-6 family, expressed on multipotent hematopoietic stem cells (HSC) and used as a marker of HSC in mice. It is found in fetal liver, adult bone marrow, peripheral blood, and spleen, and may regulate B and T cell activation.
