Target Information
CD44 cell surface antigen is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, white matter of the brain, and certain epithelial cells of the intestine and breast. Several isoforms exist, with CD44H being predominant in many normal tissues. CD44 functions as a receptor for hyaluronic acid (HA), mediating cell-cell interactions, adhesion, and migration. It plays roles in lymphocyte activation, recirculation, and homing. Expression levels may increase in certain carcinomas, potentially linked to metastatic potential. CD44 is expressed by hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia in cultured keratinocytes. Bone marrow myeloid cells and memory T cells express CD44 highly, while peripheral B and T cells can upregulate CD44 upon stimulation. The CD44 gene undergoes complex alternative splicing, producing functionally diverse isoforms. Dysfunction in CD44 is associated with diseases such as superficial keratitis and lichen sclerosus and may be implicated in tumor metastasis formation.
