Product Specific Information
- Antibody is tested in direct ELISA.
- For Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin–haptoglobin complex, protecting tissues from oxidative damage and participating in iron recycling via endocytosis and heme breakdown.
CD163 is expressed exclusively on human monocytes and macrophages, predominantly in the late phase of inflammation. It is present on all circulating monocytes and most tissue macrophages except those in mantle zones, germinal centers, interdigitating reticulum cells, and Langerhans cells. Expression is higher on CD16⁺ monocytes and upregulated by IL-10, IL-6, and dexamethasone. LPS and PMA induce shedding of CD163 into plasma or supernatant.
CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F phenotype) than dimeric haptoglobin (HP1S phenotype). It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
