Target Information
Susceptibility to tuberculosis (TB) in mice has recently been attributed to the IPR1 gene, a member of the SP100/SP140 family of nuclear body proteins. This gene encodes a leukocyte-specific nuclear body component that can function as an activator of gene transcription and may act as a nuclear hormone receptor coactivator.
Alternative splicing generates three transcript variants encoding distinct isoforms. SP110 is the closest human homolog of IPR1 and may play a role in integrating pathogen-derived signals with innate immunity and cell death mechanisms.
The IPR1/Sp110 gene product is upregulated following Mycobacterium tuberculosis infection and is required by Anaplasma phagocytophilum for infection of human promyelocytic cells. Defects in Sp110 cause severely impaired resistance to M. tuberculosis infection.
