Target Information

NLK (Nemo-like kinase) is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway, functioning downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation leads to phosphorylation of the histone methyltransferase SETDB1, forming a complex that interacts with PPARG and results in methylation and transcriptional silencing of PPARG target genes. This process inhibits adipogenesis and promotes osteoblastogenesis in mesenchymal stem cells. NLK also negatively regulates canonical Wnt/β-catenin, Notch, and MYB signaling pathways through phosphorylation-mediated inhibition of transcriptional complexes.
