Product Specific Information
The eBio1D4B monoclonal antibody reacts with mouse CD107a, also known as lysosomal-associated membrane protein-1 (LAMP-1). CD107a is a type I lysosomal membrane protein that is extensively glycosylated and constitutively expressed in late endosomes-lysosomes of all cells. It is transiently expressed on the surface of degranulating cytolytic T cells and is implicated in cancer metastasis and lysosomal storage disorders.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Verified using thioglycolate-elicited peritoneal macrophages and the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.
Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact.

Target Information
LAMP1 (CD107a) is a major lysosomal membrane protein, also found partially on the plasma membrane. It is heavily glycosylated and plays a role in lysosome biogenesis and fusion with phagosomes. LAMP1 translocates from the trans-Golgi network to endosomes and lysosomes, and upon activation, to the plasma membrane via a tyrosine-based motif (YXXI). Mutations affecting this motif alter lysosomal localization.
Cell surface LAMP1 and LAMP2 promote adhesion of PBMCs to vascular endothelium, suggesting involvement in inflammation. Increased LAMP1 expression is observed in Alzheimer’s disease, thymic involution, and mast cell activation.
