Target Information
The pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to regulate genetic programs. Several pax genes are linked to developmental mutations:
- PAX3: associated with Waardenburg syndrome
- PAX6: associated with Aniridia
- PAX2: associated with Wilms tumor
During embryogenesis, PAX2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Its expression decreases as the tubular epithelium matures. However, persistent PAX2 expression is observed in undifferentiated epithelium of Wilms’ tumors, correlating with proliferation of poorly differentiated epithelial cells. Expression of the Wilms’ tumor suppressor protein WT1 coincides with down-regulation of PAX2, suggesting that WT1 can directly repress PAX2 transcription.
