Description:
Monoclonal antibody OX108 recognizes human CD200R (OX2). CD200R is an inhibitory receptor with a novel NPXY phosphotyrosine binding domain that, upon SHIP binding, inhibits ERK, JNK, and MAPK p38 pathways, leading to suppression of TNFα secretion in activated macrophages. CD200R isoforms are involved in differentiation, particularly in tolerogenic dendritic cells.

Expression:
Restricted to hematopoietic cells — myeloid (monocytes, macrophages, DCs, neutrophils, mast cells, basophils) and subsets of T lymphocytes, Langerhans cells, and dendritic epidermal T cells. The OX108 epitope is near the CD200 binding site.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Fluorophore Details:
PerCP-eFluor® 710 emits at 710 nm, excited by blue laser (488 nm). Recommended filter: 710/50 bandpass (alternative: 695/40). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days in dark at 4°C using:

• 1-step Fix/Lyse Solution (Product #00-5333)
  Minimal impact on brightness and FRET efficiency.

Target Information

CD200R (OX2R) is a receptor for OX-2 membrane glycoprotein. Both are cell surface glycoproteins with two immunoglobulin-like domains. CD200R is mainly expressed on myeloid lineage cells and mediates downregulatory signaling. In mice, CD200R is found on monocytes, neutrophils, T-lymphocytes, and mast cells. Alternative splicing generates multiple transcript variants. Dysfunction of CD200R is associated with primary hypertrophic osteoarthropathy.
