Target Information
The serine/threonine kinase PAK is a downstream effector of small GTPases Cdc42 and Rac. PAK associates with Nck, PI3-kinase subunits (p85, p110), and PIX (PAK-interacting exchange factor) in a focal complex. PIX binding is essential for PAK localization and activation in the Cdc42–Rac signaling pathway through the SH3 domain of PIX. PIX exists as alpha and beta isoforms, both highly expressed in human and rat heart, muscle, and thymus. AlphaPIX acts as a guanine nucleotide exchange factor (GEF) on Rac, mediating lamellipodia formation, and is phosphorylated via PDGF and EphB2 receptor signaling or PI3-kinase association.
