Target Information
Ribonucleases (RNAses) H are enzymes that hydrolyze the RNA strands of RNA/DNA hybrids. Their primary role is to remove the RNA strand from RNA/DNA hybrids formed during DNA replication and repair.
RNAse H2 consists of three subunits, all necessary for enzymatic activity. Mutations in RNAse H2A or other subunits have been linked to Aicardi-Goutieres syndrome (AGS), a neurological disorder with symptoms similar to viral brain infections, including elevated IFN-alpha levels in cerebrospinal fluid.
Similar pathologies are seen with TREX1 mutations, suggesting that RNAse H2 and TREX1 have related functions. Mutations in these genes may cause intracellular nucleic acid accumulation, triggering an inflammatory response via innate immune activation.
