Target Information
Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction by generating the second messengers inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate.
There are multiple mammalian PLC isozymes, including PLCβ1, PLCβ2, PLCβ3, PLCβ4, PLCγ1, PLCγ2, PLCδ1, PLCδ2, and PLCε.
PLCδ1 functions as a calcium signal amplifier and is activated by atypical GTP-binding proteins. It acts as an effector for transglutaminase II-mediated oxytocin receptor and α1B-adrenoreceptor signaling. Mouse PLCδ1 is highly expressed in brain, heart, lung, and testis. Abnormal accumulation of PLCδ is observed in Alzheimer’s disease brains, suggesting a role in AD pathology.
PLCδ2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma. Its expression in type I intestinal metaplasia is generally benign but may evolve toward neoplastic transformation, making PLCδ2 a potential marker for gastric malignancy.
