Target Information
PROZ (Protein Z) is a vitamin K-dependent plasma glycoprotein (~62 kDa, 396 amino acids). It consists of four domains: a Gla-rich region, two EGF-like domains, and a trypsin-like domain. Although structurally related to serine proteases (factors VII, IX, X, and protein C), it lacks catalytic activity due to absence of the serine residue.
Protein Z participates in the coagulation cascade by facilitating degradation of factor Xa via the protein Z-dependent protease inhibitor (ZPI). Deficiency states have been variably associated with thrombosis or bleeding tendencies. Its activity is impaired under warfarin therapy due to vitamin K dependency.
