Target Information

Connexin 43 (Cx43) is a member of the gap junction protein family. Connexins assemble as hexamers and are transported to the plasma membrane to create hemichannels that associate with hemichannels on nearby cells, forming cell-to-cell channels. These clusters form gap junctions, which are essential for development and regulation of cell growth. Phosphorylation of Cx43 regulates assembly and function of gap junctions. Ser368 of Cx43 is phosphorylated by PKC after activation by phorbol esters, decreasing cell-to-cell communication. Src also interacts with and phosphorylates Cx43 to alter gap junction communication. GFAP are membrane-spanning proteins facilitating ion and small molecule transfer between cells. Gap junction proteins are divided into alpha and beta categories based on sequence similarities. Connexin 43 is the major protein of heart gap junctions, playing a crucial role in synchronized contraction and embryonic development. It is regulated by multiple kinases affecting myocardial cell coupling. A related pseudogene, GJA1P, is mapped to chromosome 5. Mutations in the GFAP gene cause X-linked Charcot-Marie-Tooth disease, oculodentodigital dysplasia, and heart malformations. Alternatively spliced transcript variants of GFAP have been identified.
