Product Specific Information

Description:
The SJ25C1 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells at all developmental stages except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, Leu13, and MHC class II, associating with the B-cell receptor (BCR). Signaling through CD19 induces phosphorylation, calcium flux, and B-cell proliferation. The SJ25C1 and HIB19 antibodies recognize overlapping epitopes.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Pre-titrated for use at 5 µL (0.8 µg)/test.
A test is defined as the amount of antibody required to stain a sample in a final 100 µL volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 acts as a co-receptor with the BCR to amplify antigen receptor signaling, regulating B-cell proliferation and differentiation. Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B-cell hyperactivity.
