Target Information
STK4 (MST1) is a serine/threonine kinase involved in apoptosis. It may induce chromatin condensation observed during this process. STK4 is a stress-activated, pro-apoptotic kinase that, after caspase cleavage, enters the nucleus and triggers chromatin condensation followed by DNA fragmentation.
It is a key component of the Hippo signaling pathway, crucial for organ size control and tumor suppression by limiting proliferation and promoting apoptosis. Within this cascade, STK3/MST2 and STK4/MST1 complexed with SAV1 phosphorylate and activate LATS1/2 (with MOB1), which then phosphorylate and inactivate YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 prevents its nuclear translocation, regulating genes responsible for proliferation, cell death, and migration.
STK3/MST2 and STK4/MST1 are essential for repressing proliferation of mature hepatocytes, preventing activation of adult liver stem cells (oval cells), and inhibiting tumor formation.
