Description:
CXNFT monoclonal antibody는 mouse NOS2 (inducible NOS, iNOS)에 반응합니다. Nitric oxide synthase(NOS) 효소는 L-arginine으로부터 nitric oxide를 생성하며, NOS1(nNOS)과 NOS3(eNOS)는 항상 발현되지만 NOS2는 IFN-γ, TNF-α 등 염증성 사이토카인에 의해 유도됩니다. NOS2는 M1 macrophage 및 myeloid-derived suppressor cell에서 발현됩니다.

Applications Reported:
CXNFT 항체는 세포 내 염색 후 유세포분석(Flow Cytometry)에 사용 가능합니다.

Applications Tested:
이 항체는 thioglycolate로 유도된 복막 대식세포를 이용한 세포 내 염색 실험에서 검증되었습니다. Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00)를 사용하며, 실험 조건에 따라 ≤0.125 µg/test로 사용 가능합니다.
세포 수는 10⁵~10⁸ cells/test 범위에서 최적화가 필요합니다.

Fluorochrome Information:
Alexa Fluor 700은 633–647 nm 레이저로 여기되며, 723 nm에서 방출합니다. 대부분의 장비에서는 685 LP mirror와 710/20 band pass filter가 필요합니다.

Target Information

iNOS (Inducible Nitric Oxide Synthase)는 세포 간 신호전달에 관여하는 NO 생성을 매개합니다. 간세포, 대식세포, 평활근세포 등 다양한 세포에서 발현되며, LPS나 사이토카인(IFN-γ, TNF 등)에 의해 유도됩니다.
iNOS는 L-arginine을 산화시켜 L-citrulline과 NO를 생성하며, calmodulin, NADPH, FAD, FMN 결합 도메인을 포함합니다.
iNOS 이상은 achalasia, impotence 등과 관련이 있습니다.
