Target Information
TMP21 (TMED10) is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is a component of the presenilin complex and modulates the γ-secretase, but not the ε-secretase, cleavage activity of the amyloid precursor protein (APP). The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1, which together cleave APP at γ- and ε-sites, leading to accumulation of the Aβ cleavage product associated with Alzheimer’s disease.
Co-immunoprecipitation experiments indicate that TMP21 may regulate this complex. Suppression of TMP21 expression by siRNA increases γ-secretase activity and Aβ production, showing TMP21’s role in modulating γ-secretase activity.
