The JES5-2A5 antibody reacts with mouse interleukin-10 (IL-10) and functions as a neutralizing antibody. Mouse IL-10 (18 kDa) is also known as Cytokine Synthesis Inhibitory Factor (CSIF). In mice, Th2 cells, B1 cells, macrophages, and keratinocytes produce IL-10. IL-10 inhibits Th1 cytokine synthesis and T cell proliferation, while acting as a costimulatory signal for mast cells, developing thymocytes, and the Th2 response.
Applications Reported: ELISA, ELISPOT, neutralization of IL-10 bioactivity, and intracellular staining for flow cytometric analysis.
Applications Tested: Detection antibody in sandwich ELISA for mouse IL-10, used with JES5-16E3 (14-7101) for capture and recombinant mouse IL-10 (14-8101) as standard. Recommended range: 0.5–2.0 µg/mL. Standard curve: 4000 pg/mL–30 pg/mL.
Target Information
IL-10 encodes a cytokine mainly produced by monocytes and some lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production, blocks NF-kappa B activity, and participates in JAK-STAT pathway regulation.
Knockout studies show IL-10 is essential for intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Associated diseases include Graft-Versus-Host Disease and HIV Type 1.
