Product Specific Information

This antibody recognizes an extracellular epitope of CD161, a type II transmembrane C-type lectin receptor expressed on the plasma membrane of NK cells, dendritic cells, activated monocytes, and a subset of T cells as a disulphide-linked homodimer.

Target Information

KLRB1 (CD161) is a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a small subset of CD3+ T cells. KLRB1 forms a homodimeric surface protein (40–44 kDa) and plays an inhibitory role in NK cell activity. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, leading to AKT1/PKB and RPS6KA1/RSK1 kinase activation and T cell proliferation.
KLRB1 binds to carbohydrate epitopes such as Gal-α(1,3)Gal and N-acetyllactosamine, and interacts with CLEC2D/LLT1 to inhibit NK cell-mediated cytotoxicity and interferon-gamma secretion. The protein includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
