Target Information
ERN1 (Endoplasmic Reticulum to Nucleus Signaling 1) is a serine/threonine kinase that acts as the sensor of unfolded proteins in the endoplasmic reticulum (ER). It initiates the unfolded protein response (UPR) by splicing XBP1 mRNA, converting it into a potent transcriptional activator. ERN1 is essential for fetal viability and plays a critical role under stress conditions that lead to protein misfolding in the ER. Null mutants are viable, but IRE1 is required under stress conditions. Depletion of IRE1p leads to apoptosis when unfolded proteins accumulate.
