Target Information
Neurofilaments (NFs) are a type of intermediate filament (IF) expressed almost exclusively in neuronal cells, particularly in large axons. In most vertebrates, NFs are composed of three different polypeptide chains — neurofilament medium protein (NF-M), high (NF-H), and light protein (NF-L). These share sequence and structural similarity in their coiled-coil core domains but differ in the length and sequence of their termini, especially the C-termini of NF-M and NF-H, which form flexible extensions linking NFs to each other and to other cytoskeletal elements.
NF-M protein tail-mediated interactions are critical for axonal size and cytoskeletal architecture. NF-M phosphorylation and O-GlcNAcylation are reciprocally regulated and influence its translocation, filament formation, and function. Antibodies against neurofilament subunits serve as valuable neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
