Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. Its expression is synchronous with surface IgM and plays a role in calcium conductance, cell cycle progression, and B-cell proliferation. CD20 is associated with lipid rafts, with its association modulated by extracellular stimuli and BCR aggregation. Upon receptor ligation, CD20 colocalizes with BCR before dissociation and remains on the cell surface. CD20 is a key therapeutic target for antibody-mediated B-cell depletion, as it is highly expressed on B-cell malignancies and remains surface-bound following antibody treatment. Dysfunction of CD20 is linked to Ms4a1-related common variable immune deficiency.
