Specificity Comments

Haptoglobin (Hp) is a blood plasma protein that binds free hemoglobin released from erythrocytes, inhibiting its oxidative activity. This binding sequesters iron within hemoglobin, preventing iron-utilizing bacteria from benefiting from hemolysis. Haptoglobin concentrations may increase in response to inflammation. The Haptoglobin-Hemoglobin complex is removed by the reticulo-endothelial system.

Haptoglobin consists of two α and two β chains connected by disulfide bridges. In humans, two allelic forms exist: Hp1 and Hp2, with three phenotypes (Hp1-1, Hp2-1, Hp2-2) associated with various diseases such as diabetes, cardiovascular disease, Parkinson’s, and Crohn’s disease. Haptoglobin levels are useful in diagnosing hemolytic anemia. It is expressed in hepatocytes and tissues such as skin, lung, and kidney.

Target Information

Haptoglobin is an α2-globulin plasma protein synthesized in the liver. It functions as an acute phase protein and scavenger of hemoglobin from hemolyzed red cells. The monomer consists of two α and two β chains; α chains may be hp1Fa, hp1Sa, or hp2a types, while the β chain is constant. Three major phenotypes (Hp1-1, Hp2-1, Hp2-2) are recognized.
