Target Information
The platelet-derived growth factor (PDGF) family consists of four members: PDGF-A, PDGF-B, PDGF-C, and PDGF-D. These factors form disulfide-bonded dimeric isoforms that are essential for growth, survival, and function of connective tissue cells. PDGF signaling occurs through two tyrosine kinase receptors, PDGFR-α and PDGFR-β, and is critical for organ development.
PDGF-D has a two-domain structure similar to PDGF-C and is secreted as a disulfide-linked homodimer (PDGF-DD). It induces cellular transformation and promotes tumor growth by enhancing cell proliferation and neovascularization. PDGF-D expression is found in fibroblastic adventitial cells, endothelial cells, and various tumor cell lines (ovarian, renal, lung, astrocytoma, medulloblastoma). It is also expressed in human kidney tissue. Its potential oncogenic activity may contribute to prostate cancer progression.
