Product Specific Information

The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins. D7 recognizes both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells.
Non-activated splenocytes from Ly-6.2 strains (C57BL, SJL, 129, AKR, etc.) show higher frequency of D7-positive cells compared to Ly-6.1 strains (BALB/c, C3H, NZB, etc.). Sca-1 expression increases upon activation regardless of Ly-6 haplotype.
The Ly-6 family is involved in regulation and function of T cell activation, and Sca-1 serves as a key marker for mouse hematopoietic progenitor/stem cell subsets.

Applications Tested: Flow cytometric analysis of mouse bone marrow cells (≤0.5 µg/test, 100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information

Stem Cell Antigen-1 (Sca-1) is a member of the lymphocyte antigen 6 (Ly-6) family. It is expressed on multipotent hematopoietic stem cells (HSC) and serves as a marker for murine HSC.
Sca-1 is used in combination with lineage depletion antibodies to identify murine HSC, and is found in fetal liver, adult bone marrow, peripheral blood, and spleen.
It may play a role in the regulation of B and T cell activation.
