Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 연관 키나아제에 의해 인산화되어 핵으로 이동한 후 전사 인자로 작용합니다. 이 단백질은 사이토카인과 성장인자에 반응하여 전사 활성화를 유도하며, CREB-binding protein 등의 보조활성화 인자가 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, JAK1과 JAK2에 의한 Tyr705 인산화는 STAT3의 이합체 형성, 핵 이동, DNA 결합 활성에 필수적입니다.
인간에서 STAT3 유전자는 염색체 17q에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3는 다양한 유전자 발현을 매개하며 세포 성장, 세포사멸 등 여러 세포 과정에서 중요한 역할을 합니다.
또한 Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제인자로 알려져 있습니다. STAT3는 세 가지 스플라이싱 변이체로 존재하며, STAT3 유전자의 결실은 배아 발생 초기 단계에서 치명적입니다.
