Target Information

Proteolytic degradation is essential for maintaining proper levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle control, transcription, and signaling.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins in eukaryotic cells, while the vacuolar pathway handles others through endosomes, lysosomes, and the endoplasmic reticulum.

The 26S proteasome is an ATP-dependent, multisubunit (~31) complex (~2.5 MDa) comprising a 20S proteolytic core capped by one or two 19S regulatory subunits. The 19S complexes recognize ubiquitinated proteins and mediate their unfolding and translocation into the 20S core.

An enzymatic cascade attaches multiple ubiquitin molecules to lysine residues of target proteins. Defects in the ubiquitin-proteasome pathway are linked to several genetic diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
