The M1/70.15 monoclonal antibody (mAb) reacts with the alpha-M subunit of Mac-1 (CD11b/CD18).
For this PE-Texas Red™ conjugate: excitation 488 nm / peak emission 616 nm.
Applications include immunoprecipitation, flow cytometry staining, IHC of acetone-fixed frozen sections, and blocking of CR3-mediated rosetting.

• Use dim light during handling and incubation.
• Analyze cells within 18 hours of staining.
• Dilute only the quantity to be used within one week.
• Each lot tested by flow cytometry using freshly harvested mouse blood.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule that associates non-covalently with integrin beta-2 (CD18).
The CD11b/CD18 heterodimeric complex (integrin alpha-M beta-2, Mac-1, CR3) functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
These interactions are essential for cell-cell and cell-matrix adhesion.

CD11b is expressed on:

• Monocytes/macrophages
• Granulocytes
• Activated lymphocytes
• NK cell subsets
• Dendritic cell subsets
• Microglia in the brain

Expression levels:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes
  Used as a microglial marker in nervous tissue. Expression increases during monocyte maturation and varies among macrophages. Peritoneal macrophages express higher CD11b than splenic macrophages.

Associated diseases include systemic lupus erythematosus and ITGAM-related susceptibility to lupus.
