Target Information
X-linked ectodysplasin-A2 receptor (XEDAR) is a member of the tumor necrosis factor receptor family, highly expressed during embryonic development. It binds to ectodysplatin-A2 (EDA-A2). Two major isoforms, XEDAR-s and XEDAR-L, differ by a 21-amino acid region in the cytoplasmic domain. Both isoforms lack a death domain and activate NF-kappa-B and JNK pathways via TRAF3 and TRAF6. Upon EDA-A2 treatment, cells expressing XEDAR form complexes with FADD, caspase-8, and caspase-10, leading to caspase-8 and caspase-3 activation and apoptosis. EDA-A2-induced apoptosis depends on caspase-9 activation.
