Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. DNA double-strand breaks (DSBs) in eukaryotes trigger phosphorylation of serine 139 at the C-terminus of H2AX, producing gamma-H2AX. This phosphorylation can be detected by Western blotting or immunofluorescence to assess DSB frequency.

Phosphorylation of H2AX is mediated by phosphatidylinositol 3-kinases, including ATM, DNA-PK, and ATR. H2A.X contains 143 amino acids and is synthesized during both G1 and S-phase. Its mRNA exists in two forms: a short replication-type (~575 bases, no polyA) and a long basal-type (~1600 bases, with polyA). The gene maps to chromosome 11q23.2–q23.3 in humans.

H2A.X plays a critical role in chromatin structure and DNA repair by regulating the interaction of MDC1 (mediator of DNA damage checkpoint protein 1) with DNA damage sites.
