Product Specific Information
This antibody reacts with an extracellular epitope of CD33, a 67 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed on myeloid progenitors, monocytes, granulocytes, dendritic cells, and mast cells. It is absent on platelets, lymphocytes, erythrocytes, and hematopoietic stem cells.
Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, mediating inhibitory signaling and receptor removal via ubiquitination.
CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surfaces. It is a therapeutic and diagnostic marker for acute myeloid leukemia (AML), expressed on granulocyte and macrophage precursors but not on pluripotent stem cells.
CD33 serves as a marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias. Dysfunctions of CD33 have been associated with gallbladder lymphoma and extracutaneous mastocytoma.
