Target Information
PLEKHM1은 RUN 도메인, 두 개의 Pleckstrin Homology 도메인, 그리고 Cysteine-rich 도메인을 포함하는 Pleckstrin homolog domain-containing protein M family에 속합니다.
인간에서 골화 장애(osteopetrosis)의 원인으로 확인되었으며, Rab7과 함께 후기 엔도좀/리소좀 소포에 공존합니다.
Rab7의 프레닐화가 이 공존에 필수적입니다.
돌연변이형 PLEKHM1을 가진 환자의 단핵세포는 정상적으로 osteoclast로 분화되지만, ruffled border 형성이 결여되고 골 흡수 능력이 저하됩니다.
또 다른 돌연변이는 소포 산성화를 저해하고 TRACP 분비를 증가시켜, PLEKHM1이 엔도좀 성숙 및 osteoclast-osteoblast 간 교차 신호에 중요한 역할을 함을 시사합니다.
