Target Information

Varicella Zoster Virus (VZV), a member of the human herpesvirus family, causes two distinct clinical manifestations: childhood chickenpox (Varicella) and shingles (zoster).
Varicella is the outcome of the primary infection with VZV, whereas zoster results from VZV reactivation from latently infected sensory ganglia, primarily in aging or immunosuppressed individuals.

Glycoprotein E (gE), a 623 amino acid type I integral membrane protein, is expressed on plasma membranes and in the cytoplasm of VZV-infected cells, where it likely resides in intracellular vesicles or organelles.
VZV gE forms complexes with gI in the rough endoplasmic reticulum, and most gE exists as gE–gI heterodimers in infected cells. gE is also a predominant component of the virion envelope.

The gE gene (ORF68) is essential for VZV replication. In contrast, gE genes can be deleted from herpes simplex and pseudorabies viruses, though with reduced infectivity. Since VZV lacks a gD homologue, gE (or the gE–gI complex) may perform functions typically divided between gD and gE in other alphaherpesviruses.
