Target Information
ALAS-1은 미토콘드리아 기질 내에서 글리신과 석시닐-CoA의 축합 반응을 촉매하여 δ-아미노레불리네이트, CO₂ 및 CoA를 생성하는 핵 인코딩 효소입니다. 이는 헴 생합성 경로의 첫 번째이자 속도 제한 단계에 관여하며, 헴 단백질과 시토크롬을 위한 헴을 공급합니다. ALAS1은 간에서 헴에 의해 음성 피드백 조절을 받으며, cAMP 및 호흡 해리제에 의해 전사가 유도되고, 포르볼 에스터 및 인슐린에 의해 억제됩니다. 또한 ALAS1은 헴 생합성과 생체 시계의 상호 조절에 관여하는 것으로 알려져 있으며, 생체 리듬 장애 치료의 잠재적 표적이 될 수 있습니다. 급성 간부전 시 ALAS1 발현이 감소하여 헴 대사 및 간 기능에 영향을 미칠 수 있습니다.
