Target Information
This gene generates several transcript variants differing in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode isoforms known to function as inhibitors of CDK4 kinase. Another transcript encodes an alternate open reading frame (ARF) that produces a structurally unrelated protein functioning as a stabilizer of tumor suppressor p53 by interacting with and sequestering MDM2, an E3 ubiquitin-protein ligase responsible for p53 degradation. Despite structural and functional differences, the CDK inhibitor isoforms and ARF product share a common role in cell cycle G1 control. This gene is frequently mutated or deleted in various tumors and is an important tumor suppressor gene.
